High Speed Three Dimensional Tracking of Swimming Cell by Synchronous Modulation Between TeCE Camera and TAG Lens

Abstract

This letter reports a prototype high-speed volume feedback microscope system, named a 1 ms 3D feedback microscope. In the proposed microscope system, three-dimensional information of the measured object can be acquired by synchronizing the light emission from an LED and the movement of a fast variable-focus lens. An image corresponding to a particular focal length can be measured by flashing the LED when the lens is set at a particular focal length. When LEDs of multiple colors are modulated, a color image including multiple focal planes can be obtained. We constructed a prototype of the proposed microscope system and conducted experiments to confirm its effectiveness. Experimental results showed that the proposed microscope system could quickly measure three-dimensional information of the measured object.
