Preparation of Oligopeptide Hydrogel with Rapid Hemostatic and Broad‐Spectrum Antibacterial Properties for Accelerating Wound Healing

Abstract

The development of biomaterials with rapid hemostatic and antibacterial activity is essential for the treatment of chronic bacterial infected wounds. In this study, bacterial membrane chromatography combined with one-bead-one-compound (OBOC) method is used for rapid screening of antimicrobial peptides with potential antimicrobial properties. The screened antimicrobial peptide LKAPI (AMP LI5) has broad-spectrum bactericidal properties. Besides, AMP LI5 exhibited superior activation of the coagulation cascade and increased adherence to blood cells and platelets, which allows it to quickly coagulate blood. The hemostatic results showed that the AMP LI5 has a desirable hemostatic effect on the mouse tail-breaking model (9 ± 0.47 s). Finally, an HA-base composite antimicrobial peptide hydrogel is prepared antibacterial and hemostatic effects, which have injectability, compressibility, self-healing, and good biocompatibility. In addition, the antimicrobial peptide hydrogel significantly accelerated wound healing in chronic bacterial infected wounds in mouse. In summary, antimicrobial-hemostatic peptide hydrogels are expected to be competitive and multifunctional wound dressings for hemostasis and healing of bacterial-infected wounds.
